Protein Analysis 

The main goal of the IBVF`s Protein Analysis Facility is the identification of proteins of biological interest using mass spectrometry. This Facility is available to any public or private institution on request. The analyses provided are:

• • Massive protein identification in solution by mass spectrometry coupled to high performance liquid chromatography (LC-MS/MS). Protein extracts are digested with trypsin. Peptides are subjected to high-performance liquid nanochromatography (nanoHPLC), where they are separated on a reverse phase C18 column, ionized by electrospray and fragmented in the triple quadrupole-TOF hybrid, generating a library of thousands of spectra. Protein Pilot software (Sciex) allows for identifying thousands of proteins with these MS/MS spectra. Depending on the complexity of the protein sample, three types of different gradients are offered in chromatography. Protein relative quantification (label free) by SWATH technology is also possible with this equipment.
• • In gel Protein identification. Proteins are digested with trypsin inside the acrylamide gel and tryptic peptides are extracted. Then, peptides are subjected to LC-MS/MS. The work should be carried out as clean as possible in order to prevent keratins contamination.)

The Facility has different systems for the preparation and the analysis of the samples:

• • • System for automated cutting of proteins in acrylamide gels (EXQuest Biorad).
    • Hybrid mass spectrometer, Triple quadrupole-TOF, 5600 (Sciex) Model, coupled to nanoLC, allowing LC-MS / MS analysis.
